چکیده

Abstract Background Low conception rate (CR) despite insemination with morphologically normal spermatozoa is a common reproductive restraint that limits buffalo productivity. This accounts for significant loss to the farmers and dairy industry, especially in agriculture-based economies. The immune-related proteins on sperm surface are known regulate fertility by assisting their survival performance female tract (FRT). Regardless of importance, very few studies have specifically catalogued proteome. study was designed determine identity ascertain if epididymal expressed beta-defensins (BDs), implicated male fertility, translated applied onto along other proteins. Results raw mass spectra data searched against an in-house generated proteome database from UniProt using Comet search engine identified more than 300 ejaculated which were bound either non-covalent (ionic) interactions or glycosylphosphatidylinositol (GPI) anchor. singular enrichment analysis (SEA) revealed most these extracellular varied binding activities involved immune processes. Flow cytometry six FITC-labelled lectins confirmed prediction glycosylation Several anti-microbial peptides including BuBD-129 126 also amongst presence subsequently RT-qPCR, immunofluorescence vitro fertilization (IVF) experiments. Conclusions heavily glycosylated because secreted (glyco) like BDs GPI-anchored (GPI-APs). pattern sperm-surface, however, could be perturbed elevated salt concentration incubation PI-PLC. identification numerous strengthens our hypothesis (BuBDs) assist FRT. Our results suggest sperm-surface BD role function. Further elucidating its exact physiological function required better understand regulation fertility.

mTOR, AMBRA1, and autophagy: An intricate relationship

Autophagy is the primary catabolic cellular degradation process, which generates nutrients and energy to maintain essential cellular activities upon nutrient starvation. The regulation of autophagy is central to the understanding of its mechanisms; post-translational modifications account for the rapid kinetics that control autophagy induction. A recent paper of ours provided 2 important insigh...

Crystallization of Galectin-8 Linker Reveals Intricate Relationship between the N-terminal Tail and the Linker

Galectin-8 (Gal-8) plays a significant role in normal immunological function as well as in cancer. This lectin contains two carbohydrate recognition domains (CRD) connected by a peptide linker. The N-terminal CRD determines ligand binding specificity, whereas the linker has been proposed to regulate overall Gal-8 function, including multimerization and biological activity. Here, we crystallized...

lncRNAs and MYC: An Intricate Relationship

Long non-coding RNAs (lncRNAs) are emerging as important regulators of gene expression networks, acting either at the transcriptional level, by influencing histone modifications, or at the post-transcriptional level, by controlling mRNA stability and translation. Among the gene expression networks known to influence the process of oncogenic transformation, the one controlled by the proto-oncoge...

Transcriptome Profiling Reveals Disruption of Innate Immunity in Chronic Heavy Ethanol Consuming Female Rhesus Macaques

It is well established that heavy ethanol consumption interferes with the immune system and inflammatory processes, resulting in increased risk for infectious and chronic diseases. However, these processes have yet to be systematically studied in a dose and sex-dependent manner. In this study, we investigated the impact of chronic heavy ethanol consumption on gene expression using RNA-seq in pe...
